Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the accurate detection of phosphorylated IRS-1 protein levels at the Ser636 site. IRS-1 (insulin receptor substrate 1) is a key adaptor protein involved in insulin signaling pathways, playing a pivotal role in glucose metabolism and cellular growth. Phosphorylation of IRS-1 at the Ser636 residue regulates its function and impacts insulin sensitivity, making this kit a valuable tool for research into diabetes, obesity, and other metabolic disorders.

The assay demonstrates reactivity with Human, Mouse, and Rat samples and has a dynamic range of > 5000 Cells. It employs a colorimetric detection method measured at 450 nm. The kit uses an indirect ELISA format in which IRS-1 (Phospho-Ser636) is captured by specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling HRP to catalyze a colorimetric reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Following colorimetric measurement, the Crystal Violet whole-cell staining method can be used to determine cell density, allowing results to be normalized to cell amounts. The kit is suitable for adherent cells and can be used both to measure relative amounts of phosphorylated IRS-1 in cultured cells and to screen for the effects of various treatments, inhibitors, or activators on IRS-1 phosphorylation.
