Description

Manufactured by Assay Genie, this Cell-Based ELISA Kit enables colorimetric detection of phosphorylated tyrosine 1355 on the insulin receptor (IR) in cultured cells. It is a lysate-free, high-throughput assay employing an indirect ELISA format: phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyse a colorimetric reaction upon substrate addition, read at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining method is provided to determine cell density.

The kit has a stated dynamic range of >5000 Cells and is reactive with human samples. It is intended for adherent cells and is suited to measuring the relative levels of phosphorylated IR in cultured cells, as well as screening for the effects of treatments, inhibitors (e.g., siRNA or chemicals), or activators on IR phosphorylation. The target is encoded by INSR (Gene ID: 3643; UniProt ID: P06213), a receptor tyrosine kinase in which phosphorylation at tyrosine 1355 is a post-translational modification that plays a role in cell signalling pathways governing proliferation, differentiation and apoptosis, and has been implicated in diseases including cancer, diabetes and neurological disorders.
