Description

Designed for cell-based quantification of phosphorylated Interferon-gamma Receptor alpha at tyrosine 457, this Colorimetric Cell-Based ELISA Kit from Assay Genie is a lysate-free, high-throughput assay that uses an indirect ELISA format. Phosphorylated Interferon-gamma Receptor alpha (Phospho-Tyr457) is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody; the conjugated HRP then catalyses a colorimetric reaction upon substrate addition. The assay is suited to measuring relative amounts of phosphorylated Interferon-gamma Receptor alpha in cultured cells and to screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on receptor phosphorylation.

Detection is colorimetric at 450 nm, with reactivity in Human, Mouse and Rat samples and a dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method is provided for determining cell density so that absorbance values can be normalised to cell amounts and plating differences adjusted.

The target gene is IFNGR1, the ligand-binding alpha chain of the interferon-gamma receptor, a key component of the interferon gamma signalling pathway with a role in immune regulation and inflammatory processes. Phosphorylation at tyrosine 457 modulates receptor activity, making it a useful marker for investigating interferon gamma signalling dynamics in immune and disease-related research contexts.
