Integrin beta4 (Phospho-Tyr1510)Colorimetric Cell-Based ELISA Kit

The Cell-Based ELISA Kit from Assay Genie is a colorimetric assay designed to detect phosphorylation of Integrin beta4 at tyrosine 1510 in cultured cells. The kit targets Integrin beta4 (Phospho-Tyr1510) and has been validated for reactivity with Human, Mouse and Rat samples, with a stated dynamic range of > 5000 Cells and absorbance read at 450 nm.

Integrin beta4 forms the alpha-6/beta-4 heterodimer, a receptor for laminin that plays a critical structural role in the hemidesmosome of epithelial cells. Phosphorylation at Tyr1510 is involved in cell adhesion, signal transduction and downstream signaling, and abnormalities in this modification have been associated with cancer and inflammatory disorders. The assay uses an indirect ELISA format: target phosphorylation is captured by specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colour-forming reaction upon substrate addition.

For normalisation, a monoclonal anti-GAPDH antibody is included as an internal positive control, and a Crystal Violet whole-cell staining step is provided to determine cell density and adjust for plating differences.
