Description

A cell-based colorimetric ELISA kit from Assay Genie is designed for qualitative measurement of Integrin beta4 protein levels in adherent cell cultures. The lysate-free, indirect ELISA format detects the target using Integrin beta4-specific primary antibodies paired with HRP-conjugated secondary antibodies, with a colorimetric readout at 450 nm. It is suitable for high-throughput screening of treatments, inhibitors such as siRNA or chemicals, and activators, and can measure relative Integrin beta4 expression with multiple normalization methods.

The assay is reactive with human, mouse, and rat samples and supports a dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control, and Crystal Violet whole-cell staining may be used to normalize absorbance values to cell density, correcting for plating differences. The protocol involves seeding adherent cells, blocking, overnight primary antibody incubation at 4°C, secondary antibody binding, and substrate development.

Storage is at 4°C with 6 Months stability. Integrin beta4 (ITGB4, UniProt P16144) is a laminin receptor subunit with a structural role in the hemidesmosome of epithelial cells, and is implicated in cell-matrix adhesion, migration, and diseases such as epidermolysis bullosa with pyloric atresia.
