Description

The Assay Genie Integrin beta3 (Phospho-Tyr785) Colorimetric Cell-Based ELISA Kit is designed for the qualitative detection of Integrin beta3 phosphorylated at Tyr785 in cell lysates, serum, and other samples. Using a colorimetric detection method read at 450 nm, the cell-based ELISA format is reactive with Human, Mouse, and Rat samples and has a stated dynamic range of > 5000 Cells. The assay is intended for measuring the relative amounts of phosphorylated Integrin beta3 in cultured cells, as well as screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Integrin beta3 phosphorylation.

Qualitative determination of Integrin beta3 (Phospho-Tyr785) is achieved by an indirect ELISA format. Phosphorylated Integrin beta3 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density so that absorbance values can be adjusted for plating differences.

Integrin beta3 (gene symbol ITGB3; Gene ID: 3690; UniProt ID: P05106) is an integrin protein known to participate in cell-surface-mediated signalling and cell adhesion. The protein has a molecular weight of 86,694 Da. Phosphorylation of Integrin Beta3 at Tyr785 has been implicated in conditions such as cancer and cardiovascular disorders, making it a relevant biomarker for studying these disease processes and potential therapeutic strategies.
