Description

Assay Genie offers a colorimetric, cell-based ELISA kit for the qualitative detection of Integrin beta1 phosphorylated at Threonine 789 in cultured cells. The assay uses an indirect ELISA format in which target-specific primary antibodies capture the phosphorylated protein, and HRP-conjugated secondary antibodies bound to the Fc region of the primary antibody catalyse a colorimetric reaction upon substrate addition, with absorbance read at 450 nm.

Reactive with human, mouse and rat samples and intended for adherent cells, the kit is described by Assay Genie as a convenient, lysate-free, high-throughput and sensitive assay for measuring relative levels of phosphorylated Integrin beta1 and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on Integrin beta1 phosphorylation. Because the readout is qualitative, two normalisation tools are provided: a monoclonal anti-human GAPDH antibody as an internal positive control, and a Crystal Violet whole-cell staining step to relate absorbance to cell density.

Assay Genie states a dynamic range of > 5000 cells. The target, Integrin beta1 (gene symbol ITGB1; Gene ID 3688, UniProt P05556), is an integral membrane protein that heterodimerises with an alpha-3 chain to form a receptor for extracellular-matrix proteins including fibronectin, laminin, collagen, epiligrin and thrombospondin, recognising the RGD motif present in a wide array of ligands.
