Description

A Cell-Based ELISA Kit designed for the accurate detection of phosphorylated Tyr497 levels of the IL-4 receptor (CD124) in cell lysates. Manufactured by Assay Genie, this kit offers high sensitivity and specificity for research into interleukin-4 signaling, immune regulation, and inflammation, with potential relevance to conditions such as asthma, allergies, and autoimmune disorders.

The assay uses an indirect ELISA format in which IL-4R/CD124 (Phospho-Tyr497) is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and Crystal Violet whole-cell staining may be used to relate absorbance values to cell density. The procedure is lysate-free, high-throughput, and suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on IL-4R/CD124 phosphorylation.

The kit is reactive in Human and Mouse samples, with a dynamic range of > 5000 Cells and detection by Colorimetric 450 nm. IL-4R/CD124 is encoded by the IL4R gene and functions as a receptor for interleukin-4 and interleukin-13, coupling to the JAK1/2/3-STAT6 pathway and influencing Th2 differentiation, IgE production, and chemokine and mucus production at sites of allergic inflammation. Phosphorylation at Tyr497 is associated with activation of downstream signaling cascades affecting cell proliferation and differentiation.
