Description

A cell-based ELISA kit from Assay Genie designed for the accurate detection of IL-4R (CD124) levels in cell lysates and cell culture supernatants, suitable for work with adherent cells. The kit is a lysate-free, high-throughput and sensitive assay that detects the IL-4R/CD124 protein expression profile in cells. It can be used to measure the relative amounts of IL-4R/CD124 in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on the target.

Detection is colorimetric, read at 450 nm, with a stated dynamic range of > 5000 Cells, and the kit reacts with Human and Mouse samples. It is supplied in a 96-Well Microplate format, with storage at 4°C and a stability of 6 Months. The qualitative readout is achieved through an indirect ELISA format in which IL-4R/CD124 is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colour reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density so that absorbance can be normalised to cell number.

IL-4R/CD124 is the alpha chain of the interleukin-4 receptor, a type I transmembrane protein (UniProt P24394; NCBI Gene ID 3566) with a molecular weight of 88,267 Da. It binds both interleukin-4 and interleukin-13, signals through the JAK1/2/3-STAT6 pathway, and is involved in promoting Th2 differentiation, regulating IgE production, and influencing the development of allergic and autoimmune diseases. Alternate synonyms include CD124, IL4RA and IL-4RA.
