Description

Manufactured by Assay Genie, this cell-based ELISA kit is designed to measure IL-2Ralpha (also known as CD25) protein levels in cell cultures. It provides a lysate-free, high-throughput workflow for evaluating relative IL-2Ralpha expression in adherent cells, as well as screening the impact of various treatments, inhibitors (such as siRNA or chemicals), or activators on IL-2Ralpha. The qualitative indirect ELISA format uses target-specific primary antibodies together with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

Key specifications include human reactivity, colorimetric detection at 450 nm, a dynamic range of > 5000 Cells, and storage at 4°C for 6 Months in a 96-well microplate format. Two normalization approaches are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance, and Crystal Violet whole-cell staining can be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

IL-2Ralpha is a receptor for interleukin-2, forming a non-covalent dimer with a beta subunit and, in the high- and intermediate-affinity forms, also associating with a gamma subunit. It plays a critical role in immune responses, particularly in the regulation of T cell activation and proliferation. Dysregulation of IL-2Ralpha signaling has been implicated in autoimmune diseases, allergies, and transplant rejection, making it an important biomarker for studying immune function and developing targeted therapies.
