Description

This cell-based immunoassay from Assay Genie is designed for the detection of phosphorylated IL-13Ralpha1 at tyrosine 405 in cell samples. The kit enables measurement of IL-13Ralpha1 activation levels in cell-based assays, supporting research into the interleukin-13 signalling pathway and its roles in immune regulation and inflammation. The kit is suitable for adherent cells.

Detection is performed by colorimetric readout at 450 nm using an indirect ELISA format, in which target-specific primary antibodies are bound by HRP-conjugated secondary antibodies. The stated dynamic range is > 5000 Cells, with reactivity across human, mouse and rat samples. A monoclonal GAPDH antibody is included as an internal positive control for normalising absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density. The assay is lysate-free and can be applied to measure relative phosphorylation levels in cultured cells and to screen the effects of various treatments, inhibitors or activators on IL-13Ralpha1 phosphorylation.

IL-13Ralpha1 is a receptor subunit that, together with IL4RA, forms a functional receptor for IL-13 and contributes to JAK1, STAT3 and STAT6 signalling. Phosphorylation at Tyr405 enhances downstream signalling activity, making the receptor relevant to studies of asthma, allergy and autoimmune disorders.
