The IKZF2 (Helios) Transcription Factor Activity Assay from Assay Genie is a non-radioactive DNA-binding immunoassay that quantifies DNA-binding activity of IKZF2, a zinc-finger transcription factor of the Ikaros family that plays a pivotal role in regulatory T-cell (Treg) lineage stability and immune tolerance. The microplate is pre-coated with an immobilized oligonucleotide duplex carrying the IKZF2 consensus recognition sequence; activated nuclear extracts bind to the probe and are detected with an IKZF2-specific antibody followed by HRP-conjugated secondary reagent and TMB substrate development. The kit is validated for cross-reactivity with human, mouse, and rat IKZF2 in nuclear and cell lysates, supporting immunology, autoimmune disease, transplant tolerance, and hematopoiesis research workflows conducted in vitro. Reagents should be stored at -20°C, with antibody and probe aliquots protected from repeated freeze-thaw cycles; equilibrate the plate to room temperature before running the assay. Each kit is supplied as a microplate with 96 reactions and includes pre-coated plate, diluent buffers, detection antibody, HRP conjugate, TMB substrate, stop solution, and reference standard sufficient for in vitro research use.
