Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the precise measurement of IKK-beta levels in cell lysates and tissue samples, supporting research into the NF-kB signaling pathway. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, in which IKK-beta is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

The kit is described as a convenient, lysate-free, high-throughput and sensitive assay for determining the relative expression profile of IKK-beta in cultured cells, and is suitable for screening the effects of various treatments, inhibitors such as siRNA or chemicals, or activators. It shows reactivity with human, mouse and rat samples, has a stated dynamic range of greater than 5000 cells, and is supplied in a 96-well microplate format with a storage stability of 6 months at 4°C. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density for further normalization.

The target analyte, also known as inhibitor of nuclear factor kappa-B kinase subunit beta, is encoded by the IKBKB gene (Gene ID: 3551; UniProt ID: O14920) and is a key regulator of NF-kB signaling involved in immune responses, inflammation and cell survival, with dysregulation of the pathway linked to cancer, inflammatory disorders and autoimmune conditions.
