IKK-alpha Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit enables qualitative measurement of IKK-alpha protein expression in cultured cells without the need for lysate preparation. The indirect ELISA format uses IKK-alpha-specific primary antibodies, with HRP-conjugated secondary antibodies catalysing a colorimetric reaction that is read at 450 nm. A dynamic range of > 5000 cells is reported, and the assay is suited to screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on IKK-alpha in adherent cells. Reactivity is stated for human, mouse and rat samples.

For data normalisation, the kit includes a monoclonal antibody specific for human GAPDH as an internal positive control, plus a Crystal Violet whole-cell staining step to adjust absorbance values to cell density. Storage is at 4°C for 6 months. The target, IKK-alpha (gene symbol CHUK; UniProt O15111), is a serine/threonine kinase of the IKK family that phosphorylates inhibitors of NF-kappa-B, leading to their dissociation and ultimately their degradation. Through this activity, IKK-alpha is a key regulator of the NF-kappa-B signalling pathway, influencing inflammation, immune responses and cell growth.
