Description

This Cell-Based ELISA Kit from Assay Genie is designed for the qualitative detection of IkappaB-epsilon protein expression in cultured cells. It is a lysate-free, high-throughput and sensitive assay that captures IkappaB-epsilon via target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyse a colorimetric reaction. The detection method is colorimetric, measured at 450 nm, with a dynamic range of > 5000 Cells. Reactivity has been established for Human, Mouse and Rat samples, with adherent cells as the intended sample type.

Normalization is supported through two methods: a monoclonal anti-human GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalised to cell amounts. The kit allows measurement of relative IkappaB-epsilon levels in cultured cells and screening of effects from treatments, inhibitors (such as siRNA or chemicals) or activators. Storage is at 4°C for 6 months.

IkappaB-epsilon (NF-kappa-B inhibitor epsilon, gene symbol NFKBIE) inhibits NF-kappa-B by complexing with it and trapping it in the cytoplasm, inhibiting DNA-binding of NF-kappa-B p50-p65 and p50-c-Rel complexes. It interacts with RELA, REL, NFKB1 and NFKB2. The protein is highly expressed in spleen, testis and lung, followed by kidney, pancreas, heart, placenta and brain, and is also expressed in granulocytes and macrophages. It belongs to the NF-kappa-B inhibitor family, with a chromosomal location of 6p21.1, and is found in the cytoplasm and cytosol.
