ID4 Colorimetric Cell-Based ELISA

This Cell-Based ELISA Kit from Assay Genie enables qualitative detection of ID4 protein expression directly in cultured cells without the need for lysate preparation. The assay uses a colorimetric readout at 450 nm and is intended for use with adherent cells. It is reactive with Human, Mouse and Rat samples and has a dynamic range of greater than 5000 cells.

The kit employs an indirect ELISA format in which ID4 is captured by ID4-specific primary antibodies, followed by detection with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation of target absorbance values, and a Crystal Violet whole-cell staining step is provided to determine cell density so that absorbance values can be adjusted for plating differences.

ID4 (DNA-binding protein inhibitor ID-4) is a member of the inhibitor of DNA binding (ID) protein family of basic helix-loop-helix transcription factors that can act as tumour suppressors but lack intrinsic DNA-binding activity, placing the kit within the Epigenetics and Nuclear Signaling research area. The kit is stored at 4 °C with a stability of 6 months.
