Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the qualitative detection of ICK phosphorylated at tyrosine 159 in cultured cells. The colorimetric assay, read at 450 nm, uses an indirect ELISA format in which a phospho-specific primary antibody captures the target and an HRP-conjugated secondary antibody catalyses a colorimetric reaction upon substrate addition. The kit is lysate-free and supports high-throughput measurement of relative levels of phosphorylated ICK, enabling researchers to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on ICK phosphorylation.

Validated for Human, Mouse and Rat samples, the assay has a dynamic range of > 5000 Cells and is intended for use with Adherent Cells. Because the Cell-Based ELISA is qualitative, two normalization strategies are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences.

The target protein, serine/threonine-protein kinase ICK (also known as intestinal cell kinase, hICK, Laryngeal cancer kinase 2 / LCK2, MAK-related kinase / MRK or ECO), is a 71 kDa enzyme encoded by gene ICK (Gene ID: 22858; UniProt: Q9UPZ9) that is required for ciliogenesis, phosphorylates KIF3A and regulates ciliary length as well as intraflagellar transport.
