An indirect, biotin-based enzyme immunoassay from Assay Genie for quantitative detection of human IgG-class autoantibodies directed against the zona pellucida, the glycoprotein matrix that surrounds the oocyte. Anti-zona pellucida IgG is studied in reproductive immunology and infertility research because these antibodies have been associated with unexplained female infertility, fertilization failure, and poor outcomes following assisted reproduction; they are thought to disrupt sperm binding by blocking the ZP3/ZP4 glycoprotein interactions required for fertilization.

The assay is performed on a microplate pre-coated with zona pellucida antigen. Diluted serum or plasma is added so that any patient IgG specific for the zona pellucida is captured on the solid phase, followed by a biotinylated anti-human IgG secondary antibody and a streptavidin–horseradish peroxidase reporter. After addition of TMB substrate, the reaction is stopped with an acidic stop solution and absorbance is recorded on a plate reader; signal intensity is proportional to anti-ZP IgG concentration in the sample.

Each kit from Assay Genie includes a pre-coated microwell plate, ready-to-use calibrators along with positive and negative controls, biotinylated anti-human IgG conjugate, streptavidin–HRP, concentrated wash buffer, TMB substrate, stop reagent, plate seals, and a detailed protocol insert sufficient to complete the full assay run. Unopened components should be kept refrigerated between uses and protected from light, with the conjugate and streptavidin–HRP reserved for longer-term storage at -20 °C to preserve enzymatic activity.

The procedure is intended for research use only, including clinical studies that evaluate autoantibody profiles in subfertile women, IVF failure cohorts, and investigations into the immunological contribution to recurrent pregnancy loss. The plate layout supports batch testing, with calibrators used to generate a standard curve from which unknown sample titres are interpolated.
