Human ZDHHC9 (Palmitoyltransferase ZDHHC9) ELISA Kit (HUFI05596)

Assay Genie supplies a colorimetric sandwich ELISA for quantitative measurement of human ZDHHC9 (Palmitoyltransferase ZDHHC9), an enzyme central to protein palmitoylation. The double-antibody assay is run on a pre-coated microplate with absorbance read at 450 nm, and has a stated sensitivity of 46.875 pg/mL with a working range of 78.125-5000 pg/mL.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity has been assessed at 1:2, 1:4 and 1:8 dilutions, with serum (n=5) ranging 82-104%, EDTA plasma (n=5) 81-104%, and heparin plasma (n=5) 82-105%. Brief sample handling described by the manufacturer: serum is clotted and centrifuged at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C; tissue is homogenised in PBS with protease inhibitors; cell culture supernatant is clarified at 2500 rpm for 5 minutes; and cell lysates are prepared with lysis buffer containing protease inhibitors. The procedure includes primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes at 37°C, HRP-Streptavidin (SABC) for 30 minutes at 37°C, TMB colour development in the dark for 10-20 minutes, and stop-solution reading at 450 nm.
