Description

Developed by Assay Genie for the quantitative measurement of human ZAP70, this Sandwich ELISA kit uses a double-antibody format with colorimetric detection. Reactivity is human, and the assay covers a measurable range of 0.313–20 ng/mL with a sensitivity of 0.188 ng/mL. It accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with all reagents stored at 2-8°C for 12 months.

ZAP70 is a critical kinase involved in T cell signal transduction and immune system function, and its dysregulation has been linked to autoimmune disorders and certain types of leukemia. The protocol proceeds through reagent equilibration, primary incubation at 37°C for 90 minutes for antigen binding, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after adding stop solution.

Linearity has been validated in serum, EDTA plasma, and heparin plasma across 1:2, 1:4, and 1:8 dilutions (n = 5). Recovery ranges are 85–103% in serum (average 97%), 85–102% in EDTA plasma (average 93%), and 89–105% in heparin plasma (average 97%).
