Human WNT10B ELISA Kit

Manufactured by Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of human WNT10B in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a double-antibody sandwich ELISA format with colorimetric detection read at 450 nm, and is intended for research applications in metabolism, including studies of the Wnt signaling pathway and its role in conditions such as cancer, osteoporosis, and developmental disorders.

Assay performance is characterised by a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Linearity has been demonstrated across serum (n = 5) with recoveries of 86–105% at 1:2, 92–105% at 1:4, and 85–100% at 1:8; EDTA plasma (n = 5) at 82–98%, 85–98%, and 92–99% for the same dilutions; and heparin plasma (n = 5) at 82–93%, 81–99%, and 80–98%. Spike-and-recovery testing yielded ranges of 87–105% (average 96%) for serum, 86–103% (average 92%) for EDTA plasma, and 88–105% (average 98%) for heparin plasma.

The kit should be stored at 2–8 °C for up to 12 months. The assay procedure involves plate and reagent equilibration, a 90-minute primary incubation at 37 °C, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution.
