Human WNT1 ELISA Kit

A sandwich double-antibody ELISA from Assay Genie with colorimetric detection, used to quantify human WNT1. The assay is reactive with human serum, plasma, cell culture supernatant and cell or tissue lysate samples, and is assigned to metabolism research.

Reported assay specifications are a sensitivity of 7.5 pg/mL and a working range of 12.5–800 pg/mL. Dilutional linearity at 1:2, 1:4 and 1:8 is 90–103%, 90–93% and 82–100% for serum, 89–99%, 86–96% and 80–91% for EDTA plasma, and 82–100%, 87–98% and 92–103% for heparin plasma. Spiking recovery is reported as 89–102% (average 95%) for serum, 85–103% (average 98%) for EDTA plasma and 88–100% (average 94%) for heparin plasma. Storage is 2–8°C for 12 months.

The protocol involves equilibrating reagents and the pre-coated microplate to room temperature, a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, 30 minutes of HRP-streptavidin binding, TMB substrate development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after stop solution is added. Included are a pre-coated microplate, lyophilized standard, biotin-labelled antibody concentrate, HRP-streptavidin SABC, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer, plate sealers and a technical manual.
