Description

Assay Genie's sandwich ELISA kit is designed for the quantitative measurement of human Wilms tumor protein (WT1) in human serum, plasma, tissue homogenates, cell culture supernatants, and other biological fluids, using a colorimetric detection method. Reactivity is for human samples and specificity covers natural and recombinant human Wilms tumor protein.

WT1 is a DNA-binding transcription factor containing four C2H2-type zinc-finger motifs at the C-terminus and a proline/glutamine-rich DNA-binding domain at the N-terminus. It recognizes and binds to the DNA sequence 5'-GCG(T/G)GGGCG-3', regulates the expression of numerous target genes including EPO, and plays an essential role in urogenital system development. The protein exhibits both tumor suppressor and oncogenic roles in tumor formation. The target is also known by the synonym WT33 and is catalogued under UniProt entry P19544, with isoform 4 localized to the nucleus/nucleoplasm.

Performance specifications include a detection range of 15.6-1000pg/mL, sensitivity of 10pg/ml, intra-assay CV of 4.6%, and inter-assay CV of 8.3%. Recovery averages 89% (range 83-95%) in serum and 91% (range 85-97%) in plasma. Linearity (N=5) at 1:2, 1:4, 1:8, and 1:16 dilutions was 110-120%, 92-102%, 89-99%, and 104-116% respectively in serum; 94-103%, 91-101%, 104-113%, and 93-106% in EDTA plasma; and 93-102%, 92-101%, 96-105%, and 94-107% in heparin plasma. For research use only in cancer research applications.
