Description

A sandwich ELISA designed by Assay Genie for the quantitative detection of WIF1 (Wnt Inhibitory Factor 1) in human samples. WIF1 is a secreted protein that functions as a negative regulator of the Wnt signaling pathway, which plays a pivotal role in development, tissue homeostasis, and disease progression. The kit supports research into the role of WIF1 in cancer, stem cell regulation, and tissue regeneration, helping researchers investigate the Wnt pathway and its implications in disease.

The assay has a sensitivity of 18.75 pg/mL and a measuring range of 31.25–2000 pg/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and should be stored at 2–8°C for 12 months. Reported linearity across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions falls within 80–105%, and recovery spans 85–103% with averages of 94% (serum), 90% (EDTA plasma) and 95% (heparin plasma).

The protocol begins by equilibrating reagents and loading standards, samples, and controls onto the pre-coated plate, followed by incubation at 37°C for 90 minutes. A biotin-labelled detection antibody is added and incubated at 37°C for 60 minutes, then HRP-Streptavidin is applied for 30 minutes. TMB substrate is added and developed in the dark for 10–20 minutes before the stop solution is introduced and absorbance is read at 450 nm.
