Human Von Hippel Lindau/VHL ELISA Kit

From Assay Genie, this Human Von Hippel-Lindau (VHL) ELISA Kit is a sandwich ELISA, double antibody assay designed to measure levels of VHL protein in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric detection method and is intended for signal transduction research. VHL is a tumor suppressor gene with key roles in regulating cell growth, division, and survival, and mutations in the VHL gene are associated with von Hippel-Lindau disease and various other cancers.

Linearity was 86–105% (1:2), 89–105% (1:4) and 89–104% (1:8) in serum (n=5); 82–97%, 87–95% and 85–100% in EDTA plasma (n=5); and 86–98%, 83–100% and 81–94% in heparin plasma (n=5). Recovery averaged 92% in serum (range 87–99%), 99% in EDTA plasma (range 91–103%) and 95% in heparin plasma (range 87–104%). The assay procedure includes primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB color development for 10–20 minutes in the dark, and absorbance reading at 450 nm after stop solution addition.

Sample preparation protocols cover serum (centrifuge at 1000 × g for 20 minutes), plasma (centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysate.
