Description

From Assay Genie, this sandwich ELISA kit uses a double-antibody format with colorimetric detection for the quantification of vitronectin in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Vitronectin is a glycoprotein involved in cell adhesion, spreading, migration, and tissue remodelling, with relevance to wound healing, inflammation, and cancer progression, making the kit useful in cell biology research.

Performance characteristics include a sensitivity of 5.625 ng/mL and an assay range of 9.375–600 ng/mL. Linearity in serum (n = 5) was 87–104%, 90–103%, and 91–102% at 1:2, 1:4, and 1:8 dilutions respectively. EDTA plasma (n = 5) gave 83–98%, 85–100%, and 82–99%, and heparin plasma (n = 5) gave 81–99%, 82–90%, and 85–97%. Average recovery was 92% in serum (range 85–100%), 99% in EDTA plasma (range 88–105%), and 96% in heparin plasma (range 85–105%). The target protein is catalogued under UniProt P04004. The kit should be stored at 2–8°C for 12 months.

The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, then incubating at 37°C for 90 minutes. Biotin-labelled detection antibody is added and incubated for 60 minutes, followed by HRP-Streptavidin for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution.
