Description

A Sandwich ELISA from Assay Genie designed for the quantitative measurement of Vitamin K-dependent protein Z (PROZ) in human samples. The assay uses a colorimetric detection method and is suitable for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Specificity has been demonstrated for both natural and recombinant human PROZ.

The stated detection range is 0.156–10 ng/mL with a sensitivity of 0.034 ng/mL. Intra-assay CV is 6.5% and inter-assay CV is 9.8%. Recovery was assessed in serum (average 102%, range 96–108%) and plasma (average 104%, range 98–110%). Dilutional linearity (N=5) across 1:2, 1:4, 1:8 and 1:16 gave recoveries of 86–96%, 106–117%, 108–119% and 116–126% for serum; 100–109%, 102–114%, 102–112% and 106–116% for EDTA plasma; and 108–119%, 105–117%, 110–119% and 103–113% for heparin plasma, respectively.

PROZ is a liver-synthesised, secreted vitamin K-dependent glycoprotein involved in blood coagulation. It assists hemostasis by binding thrombin and, in the presence of SERPINA10, calcium and phospholipids, inhibits activated factor X. Deficiencies in PROZ have been associated with an increased risk of ischemic arterial diseases and fetal loss. For research use only.
