Description

A sandwich ELISA, double antibody kit from Assay Genie is designed for the quantitative measurement of human versican (VCAN) using a colorimetric detection method. The assay is reactive with human samples and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The manufacturer reports a sensitivity of 0.938 ng/mL and a detection range of 1.563–100 ng/mL. The kit targets versican core protein (UniProt P13611), an extracellular matrix proteoglycan that contributes to cell adhesion, migration, and tissue development, and whose dysregulation has been linked to cancer, cardiovascular diseases, and inflammatory disorders.

The procedure involves equilibrating reagents and TMB substrate to room temperature, then loading standards, samples, and controls onto the pre-coated plate. Primary incubation is carried out at 37 °C for 90 minutes, followed by biotin-labelled detection antibody binding for 60 minutes and HRP-streptavidin binding for 30 minutes. Colour is developed with TMB substrate in the dark for 10–20 minutes, the reaction is stopped, and absorbance is read at 450 nm. Reported linearity for serum at 1:2, 1:4, and 1:8 dilutions is 82–96%, 84–99%, and 80–99% respectively; for EDTA plasma, 89–98%, 88–104%, and 89–100%; and for heparin plasma, 89–100%, 83–98%, and 83–90%. Recovery ranges are 87–105% (average 94%) for serum, 85–105% (average 94%) for EDTA plasma, and 89–99% (average 96%) for heparin plasma. Unopened components should be stored at 2–8 °C for 12 months.
