Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of human VASPIN (visceral adipose-specific serine protease inhibitor) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody sandwich format with colorimetric detection, and absorbance is read at 450 nm following TMB substrate development.

Vaspin, also known as SERPINA12 or Serpin A12, is a serine protease inhibitor implicated in regulating metabolic processes and insulin sensitivity, and is studied as a biomarker in obesity-related disorders, type 2 diabetes, and cardiovascular disease. The six-step protocol comprises: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotinylated detection antibody binding at 37°C for 60 minutes; HRP-streptavidin binding at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and stop reaction followed by absorbance measurement at 450 nm.

Performance specifications include a sensitivity of 18.75 pg/mL and an assay range of 31.25–2000 pg/mL. Linearity for serum (n=5) is 88–102% at 1:2, 87–105% at 1:4, and 86–95% at 1:8; EDTA plasma (n=5) shows 82–99%, 86–101%, and 87–100%; heparin plasma (n=5) shows 80–95%, 85–100%, and 81–98%. Recovery averages are 90% for serum, 92% for EDTA plasma, and 95% for heparin plasma. Kit components include a pre-coated dismountable microplate, lyophilized standard, biotin-labelled antibody, HRP-streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer, and a technical manual. Storage is at 2–8°C for 12 months.
