Description

This sandwich ELISA Kit from Assay Genie is designed for the quantitative detection of human V(D)J recombination-activating protein 1 (RAG1), also known by the synonyms RING finger protein 74, RAG-1, and RNF74. The assay is validated for use with human samples, including serum, plasma, tissue homogenates, cell culture supernatants, and other biological fluids, and shows specificity toward natural and recombinant human RAG1.

Performance characteristics include a detection range of 0.156–10 ng/mL and a sensitivity of 0.098 ng/mL, with intra-assay precision of 4.1% and inter-assay precision of 7.3%. Recovery in serum averaged 100% (range 94–106%) and in plasma averaged 102% (range 96–108%). Linearity was assessed across 1:2, 1:4, 1:8, and 1:16 dilutions in serum, EDTA plasma, and heparin plasma, with values generally falling within the 80–122% range depending on matrix and dilution.

RAG1 is the catalytic component of the RAG complex, a multiprotein assembly that mediates the DNA cleavage phase of V(D)J recombination, a process essential for assembling immunoglobulin and T-cell receptor gene diversity in developing B- and T-lymphocytes. The protein localizes to the nucleus and is implicated in immune deficiencies and autoimmune diseases when dysregulated. The kit is for research use only (UniProt: P15918).
