Description
A sandwich ELISA from Assay Genie provides quantitative measurement of utrophin in human samples. Utrophin plays a role in muscle function and regeneration, and its expression is relevant to neuromuscular disorders including muscular dystrophy, making it a useful biomarker for studying these conditions and exploring therapeutic approaches. The kit uses a double-antibody format with colorimetric detection.
Performance specifications include a sensitivity of 0.469 ng/mL and a detection range of 0.781-50 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity in serum (n=5) ranges from 86-105% at 1:2, 85-95% at 1:4 and 94-104% at 1:8 dilution. Recovery in serum (n=5) ranges from 90-105% with an average of 98%. The kit is stored at 2-8°C for 12 months.
The assay protocol involves equilibrating reagents and the TMB substrate to room temperature, loading standards, samples and blanks, and incubating at 37°C for 90 minutes. Biotin-labeled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is added and incubated in the dark for 10-20 minutes, and absorbance is read at 450 nm after the stop solution is added.
