Description

A colorimetric, sandwich ELISA with a double-antibody format from Assay Genie, designed for the quantitative measurement of uteroglobin (also known as Clara cell protein; synonyms include CC16, CC10, CCPBP, CCSP, Clara, PCB-BP, SCGB1A1, UGB and blastokinin) in human samples. The kit is reactive with human samples and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid sample types.

Manufacturer-reported performance characteristics include a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions is given as 87–105%, 84–99% and 81–99% for serum (n = 5); 90–105%, 84–99% and 91–99% for EDTA plasma (n = 5); and 87–100%, 88–99% and 85–99% for heparin plasma (n = 5). Recovery (n = 5) averages 94% in serum (range 90–97%), 96% in EDTA plasma (range 90–104%) and 93% in heparin plasma (range 87–100%). The procedure consists of equilibrating reagents and TMB substrate to room temperature; primary incubation at 37 °C for 90 minutes; biotinylated detection antibody for 60 minutes at 37 °C; HRP-Streptavidin (SABC) for 30 minutes at 37 °C; TMB colour development in the dark for 10–20 minutes; and absorbance reading at 450 nm after addition of stop solution.

Components supplied include a dismountable ELISA microplate (pre-coated), lyophilised standard, biotin-labelled antibody (100× concentrate), HRP-Streptavidin conjugate (SABC, 100×), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25× wash buffer, plate sealers and a technical manual. The kit is stored at 2–8 °C for 12 months, with the microplate strips and lyophilised standard additionally storable at −20 °C for 12 months in sealed foil bags with desiccant.
