Description

A sandwich ELISA developed by Assay Genie for the quantitative measurement of human Uromodulin (UMOD), also known as Tamm-Horsfall urinary glycoprotein or THP, in human biological samples including serum, plasma, urine, tissue homogenates, cell culture supernates and other biological fluids. The assay uses a colorimetric detection method and is reactive with human samples, with specificity for natural and recombinant human Uromodulin.

The assay delivers a detection range of 0.156-10 ng/mL and a sensitivity of 0.078 ng/mL. Precision is reported as an intra-assay CV of 6.3% and an inter-assay CV of 10.5%. Linearity has been established in serum (99-108% at 1:2, 86-96% at 1:4, 93-103% at 1:8, 98-107% at 1:16), EDTA plasma (90-101%, 108-118%, 100-112%, 100-109%) and heparin plasma (80-92%, 91-101%, 90-102%, 99-108%). Recovery averages 96% (range 90-102%) in serum and 98% (range 92-104%) in plasma.

Uromodulin is the most abundant protein in normal mammalian urine and, in its secreted form, may contribute to colloid osmotic pressure, retard passage of positively charged electrolytes, help prevent urinary tract infection, and inhibit formation of liquid containing supersaturated salts and subsequent salt crystals. It is secreted as a homodimer that polymerizes into long filaments and is detected in urine. Defects in UMOD are linked to familial juvenile hyperuricemic nephropathy type 1 (HNFJ1), medullary cystic kidney disease type 2 (MCKD2), and glomerulocystic kidney disease with hyperuricemia and isosthenuria (GCKDHI). Uniprot accession: P07911. For research use only.
