Human UGCG ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative detection of UGCG (UDP-Glucose Ceramide Glucosyltransferase) in human samples. UGCG plays a central role in glycosphingolipid biosynthesis, contributing to cell signaling, membrane trafficking, and cell–cell interactions, and its dysregulation has been implicated in cancer, diabetes, and neurodegenerative disorders. The kit uses a double-antibody sandwich format with a colorimetric TMB readout measured at 450 nm.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a sensitivity of 0.469 ng/mL and a working range of 0.781–50 ng/mL, with kit components stable at 2–8°C for 12 months. Reported linearity at 1:2, 1:4, and 1:8 dilutions is 94–101% / 86–105% / 86–103% for serum (n=5), 86–101% / 82–101% / 82–98% for EDTA plasma (n=5), and 80–94% / 90–96% / 89–99% for heparin plasma (n=5). Recovery ranges from 85–102% (mean 92%) in serum, 86–99% (mean 94%) in EDTA plasma, and 85–104% (mean 94%) in heparin plasma.

The protocol follows a sandwich ELISA workflow: equilibrate reagents and the pre-coated plate; incubate standards and samples at 37°C for 90 minutes; add biotinylated detection antibody and incubate for 60 minutes; add HRP-Streptavidin (SABC) for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; and read absorbance at 450 nm after adding stop solution.
