Description

Assay Genie provides a high-quality sandwich ELISA kit developed for the precise quantification of Uncoupling Protein 1 (UCP1) in human samples. UCP1 is a key mitochondrial protein involved in thermogenesis and energy expenditure and plays a central role in brown adipose tissue biology, making this kit relevant to research into metabolism, energy balance, and obesity-related disorders.

The assay uses a colorimetric, double-antibody sandwich ELISA format and is reactive with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 46.875 pg/mL and the working range is 78.125–5000 pg/mL. Kit components are stored at 2–8°C for 12 months. Reported linearity in serum (n=5) spans 94–105% (1:2), 82–100% (1:4) and 87–98% (1:8); in EDTA plasma (n=5) 85–105%, 85–101% and 86–99%; and in heparin plasma (n=5) 87–99%, 87–101% and 80–100%. Recovery averages are 90% for serum (range 86–96%), 95% for EDTA plasma (86–105%) and 92% for heparin plasma (85–105%). The procedure involves reagent equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition.
