The UBN1 target, Ubinuclein-1, is a nuclear histone chaperone (~84 kDa) that partners with HIRA, CABIN1, and ASF1a to drive replication-independent deposition of the H3.3 histone variant at active promoters, enhancers, and sites of DNA damage. Its conserved HUH domain binds the H3.3-H4 dimer, and loss of UBN1 disrupts chromatin organization, embryonic stem cell identity, and senescence-associated heterochromatin foci formation.

Investigators quantifying endogenous Ubinuclein-1 levels in cell lysates or biological samples apply this Assay Genie microplate immunoassay in in vitro studies of epigenetic regulation, chromatin remodeling, and tumor biology, loading 100 µL of prepared sample per well alongside a recombinant UBN1 standard curve.
