Human Ubiquitin-conjugating enzyme E2 A (UBE2A) ELISA Kit

Designed for the quantitative measurement of human Ubiquitin-conjugating enzyme E2 A (UBE2A) in research samples, this sandwich ELISA from Assay Genie is validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with specificity for natural and recombinant human UBE2A. The assay delivers a detection range of 0.156-10 ng/mL and a sensitivity of 0.078 ng/mL.

UBE2A is a ubiquitin-conjugating enzyme that accepts ubiquitin from the E1 complex and catalyses its covalent attachment to other proteins. In association with the E3 enzyme BRE1 (RNF20 and/or RNF40), it contributes to transcriptional regulation by monoubiquitinating histone H2B at 'Lys-120' to form H2BK120ub1, a tag linked to epigenetic transcriptional activation, elongation by RNA polymerase II, telomeric silencing and prerequisite marks such as H3K4me and H3K79me. In vitro the enzyme also catalyses 'Lys-11', as well as 'Lys-48'-linked polyubiquitination, and it is required for postreplication repair of UV-damaged DNA. Dysregulation of UBE2A activity has been linked to cancer, neurodegenerative disorders and inflammatory conditions, so accurate quantification can support studies of disease mechanisms and potential therapeutic targets.

Reported intra-assay precision is 4.1% CV and inter-assay precision is 7.5% CV. Linearity has been characterised for serum, EDTA plasma and heparin plasma across dilutions of 1:2, 1:4, 1:8 and 1:16, with average recovery of 96% (range 90–102%) in serum and 98% (range 92–104%) in plasma.
