Human UBE3A ELISA Kit

This Sandwich ELISA Kit from Assay Genie is intended for the quantitative measurement of human UBE3A (ubiquitin protein ligase E3A) using a double-antibody, colorimetric detection format. The assay is configured for human samples, with serum and plasma as the principal validated matrices; the manufacturer's specifications additionally list cell culture supernatant, cell or tissue lysate, and other liquid sample types. UBE3A has been linked to protein degradation and, when dysregulated, to neurodevelopmental disorders such as Angelman syndrome as well as certain cancers.

Reported analytical sensitivity is 0.188 ng/mL, with a working assay range of 0.313–20 ng/mL. The kit is stored at 2–8°C and is stated to remain stable for 12 months under those conditions. Across 1:2, 1:4 and 1:8 dilutions, linearity is reported as 86–105% for serum, 90–105% for EDTA plasma and 85–99% for heparin plasma (n = 5). Spiking recovery is 86–103% for serum (average 95%), 88–105% for EDTA plasma (average 94%) and 87–100% for heparin plasma (average 93%). The procedure comprises six stages: equilibrate reagents and the TMB substrate to room temperature and load standards, samples and a zero control onto the pre-coated plate; incubate at 37°C for 90 minutes for antigen binding; add a biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop colour with TMB substrate for 10–20 minutes in the dark; then add stop solution and read absorbance at 450 nm.
