Human Tyrosinase ELISA Kit

Assay Genie provides a sandwich ELISA, double antibody kit for the quantitative measurement of tyrosinase in human samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The assay uses a colorimetric detection method with TMB substrate and absorbance read at 450 nm. According to the manufacturer, the kit has a sensitivity of 46.875 pg/mL and a detection range of 78.125–5000 pg/mL. Tyrosinase is described as a key enzyme in melanin production, the pigment responsible for skin, hair, and eye colour, with dysregulation linked to conditions such as vitiligo, melanoma and other pigmentation disorders.

Reported linearity values across serum, EDTA plasma and heparin plasma fall within 80–105% at dilution factors of 1:2, 1:4 and 1:8. Recovery ranges are 91–103% (serum, average 100%), 90–103% (EDTA plasma, average 96%) and 89–103% (heparin plasma, average 98%). The workflow involves reagent equilibration, a 90-minute primary incubation at 37 °C, biotinylated detection antibody incubation for 60 minutes at 37 °C, HRP-streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction prior to absorbance measurement. Kit components include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-streptavidin conjugate (SABC, 100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer and a technical manual. The kit is stored at 2-8 °C for 12 months.
