For laboratories profiling thiol-disulfide redox homeostasis in human disease models, the Assay Genie quantitative ELISA kit delivers measurement of TXNDC11, an endoplasmic reticulum–resident thioredoxin-family oxidoreductase. The protein partners covalently with the mannosidase EDEM2 through a C692 disulfide bond in its Trx5 domain, initiating the first N-glycan trimming step that commits misfolded glycoproteins to ER-associated degradation (ERAD). It is also transcriptionally upregulated by the IRE1-sXBP1 arm of the unfolded protein response and accelerates turnover of ER-resident transcription factors including ATF6α, CREB3, and CREB3L2.

The colorimetric microplate format supplies pre-coated wells, calibrated recombinant standards, biotinylated detection antibody, and substrate reagents for 96 Assays per run. Researchers commonly apply the assay to clarified cell lysates and tissue homogenates from ER-stress-challenged cell cultures. Standard curve fitting is performed against recombinant human TXNDC11 calibrators for analyte quantification. Each lot is validated to confirm specificity for the human target, with minimal cross-reactivity observed against closely related thioredoxin-domain family members under recommended running conditions.

Unopened components should be kept refrigerated and shielded from light until the printed lot expiration date. Bring working reagents to room temperature before use, and avoid repeated freeze-thaw cycles of diluted standards to preserve immunoreactivity. The kit is intended for research use only and supports programs in redox biology, ER-stress signaling, cancer cell line characterization, and neurodegeneration studies where TXNDC11 abundance is being mapped against pharmacological ERAD perturbation.
