Human Tryptophan 5-hydroxylase 1 (TPH1) ELISA Kit

A sandwich ELISA from Assay Genie for the quantitative measurement of human Tryptophan 5-hydroxylase 1 (TPH1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. TPH1 is the rate-limiting enzyme in the biosynthesis of serotonin, catalysing the biopterin-dependent monooxygenation of tryptophan to 5-hydroxytryptophan, and its dysregulation has been implicated in mental health disorders such as depression and anxiety, as well as in gastrointestinal disease. The target is the TPH1 homotetramer encoded at chromosomal location 11p15.3-p14 (UniProt P17752).

The colorimetric assay is specific for natural and recombinant human TPH1, with a detection range of 0.312–20 ng/mL and a sensitivity of 0.11 ng/mL. Reported precision is 3.6% intra-assay CV and 6.5% inter-assay CV. Linearity in serum (n=5) is 92–105% at 1:2, 100–113% at 1:4, 103–112% at 1:8 and 91–102% at 1:16, with comparable dilution profiles in EDTA and heparin plasma matrices. Average recovery is 82% in serum (range 80–88%) and 84% in plasma (range 80–90%). For research use only.
