Human Trypsinogen Activation Peptide,TAP (TAP) ELISA Kit

A Sandwich ELISA from Assay Genie for the quantitative detection of human Trypsinogen Activation Peptide (TAP) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay recognises both natural and recombinant human TAP.

The kit provides a detection range of 0.78-50 ng/mL with a sensitivity of 0.444 ng/mL. Intra-assay precision is 4.1% CV and inter-assay precision is 5.5% CV. Linearity was assessed at sample dilutions of 1:2, 1:4, 1:8 and 1:16. In serum (N=5) recoveries were 85-95%, 105-115%, 105-117% and 115-125%; in EDTA plasma (N=5) 80-90%, 100-110%, 103-103% and 89-99%; and in heparin plasma (N=5) 83-95%, 95-107%, 118-118% and 96-105%. Average recovery was 86% (range 80-92%) for serum and 88% (range 82-94%) for plasma.

Trypsinogen-activation peptide is a small peptide released when trypsinogen is activated to trypsin. Under physiological conditions, activation takes place in the intestinal lumen and is catalysed by enteropeptidase. In pancreatitis, trypsinogen is prematurely activated within pancreatic acinar cells, releasing TAP into the peripheral circulation. Although TAP is rapidly excreted via the kidneys with a circulating half-life of less than 8 minutes, significant increases in plasma and urine TAP concentrations have been reported in acute pancreatitis.
