The TRPV6 protein is a highly selective calcium ion channel belonging to the vanilloid subfamily of transient receptor potential channels. Expressed predominantly along the apical membrane of enterocytes in the duodenum and jejunum, TRPV6 mediates the rate-limiting step of transcellular calcium absorption and is therefore central to whole-body calcium homeostasis. Its expression is tightly regulated by 1,25-dihydroxyvitamin D3, the active form of vitamin D, which links dietary calcium uptake directly to endocrine signaling. Beyond the intestine, TRPV6 is also detected in placental trophoblasts, prostate epithelium, salivary gland ducts, and certain cancer cell lines, where altered channel activity has been associated with prostate tumor progression, breast cancer proliferation, and placental calcium transfer to the developing fetus.
