Quantitative measurement of soluble Triggering Receptor Expressed on Myeloid Cells 1 (sTREM-1) underpins a growing body of research into innate immune amplification, where this immunoglobulin superfamily receptor is studied on neutrophils and monocytes as a driver of acute and chronic inflammation. This Assay Genie kit is configured for research use only, providing an absorbance-based immunoassay built around paired anti-human TREM-1 antibodies that recognise both the shed ectodomain and recombinant forms of the receptor.

The assay is intended for conditioned media from cultured myeloid cell models, including monocyte-derived macrophages and neutrophil-like lines such as HL-60 and differentiated THP-1 cultures, as well as blood-derived specimen types collected under institutional review board-approved protocols. Each kit is supplied with a pre-coated microplate, calibrated standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer, substrate, and stop solution, with a standard curve sufficient for interpolating unknowns in a routine plate-reader workflow.

Investigators working on sepsis, systemic inflammatory response syndrome, acute respiratory distress syndrome, and bacterial or fungal infection have used sTREM-1 as a candidate biomarker because the soluble ectodomain is released into circulation during myeloid cell activation. Additional applications include profiling TREM-1 pathway activity in tumor-associated macrophages, characterising amplification loops in inflammatory bowel disease and rheumatoid arthritis synovial inflammation, and tracking myeloid responses to lipopolysaccharide or other TLR ligand challenges in cell culture.

Kit components are supplied refrigerated and should be stored cold upon arrival; reconstituted standards and detection antibody should be handled according to the accompanying certificate of analysis and aliquoted to limit repeated freeze-thaw cycles. The kit is provided for laboratory research use and is not cleared for clinical diagnostic decision-making.
