Quantitative measurement of human Transient Receptor Potential Melastatin 1 (TRPM1) protein in tissue homogenates and related sample matrices. This chromogenic immunoassay from Assay Genie is supplied as a complete kit providing reagents for 96 individual determinations and is intended for research use only. TRPM1 is a calcium-permeable non-selective cation channel best characterized in retinal ON bipolar cells, where loss-of-function mutations underlie complete congenital stationary night blindness. The channel is also expressed by cutaneous melanocytes, and progressive loss of TRPM1 expression has been documented during metastatic progression of melanoma, making it a focus in pigment cell biology and cutaneous oncology research. Investigators use this assay to compare TRPM1 levels across patient-derived tissue specimens, melanocyte cell line models, or experimental perturbations of intracellular calcium signaling. Sample preparation involves homogenization of flash-frozen tissue in lysis buffer, with cleared supernatants aliquoted and stored frozen until the assay run.
