Description

Verified against the Assay Genie product datasheet, this human TRPC6 immunoassay pairs a biotin-streptavidin-HRP detection chain with TMB substrate development, terminated by an acidic stop solution before absorbance is read. The reconstituted standard curve covers 0.312, 0.625, 1.25, 2.5, 5, 10, and 20 ng/mL, supporting a quantitation range of 0.312-20 ng/mL and a lower limit of detection near 0.129 ng/mL. Unopened kits carry a 12 month shelf life, with the lyophilised standard reconstituted in the supplied buffer, aliquoted into single-use vials, and stored frozen at -20 degrees C to minimise freeze-thaw drift, while working dilutions of detection antibody and streptavidin-HRP are kept refrigerated between runs.

Beyond podocyte and focal segmental glomerulosclerosis research, the kit supports investigation of TRPC6 in HEK293 and vascular smooth muscle overexpression systems, idiopathic pulmonary arterial hypertension patient-derived lung homogenates, and pressure-overload cardiac hypertrophy tissue cross-checked against human samples where channel expression is being benchmarked. The capture antibody is human-specific, with no significant cross-reactivity observed against rat or mouse TRPC6 under standard conditions, so rodent lysates should be validated with a species-matched assay if quantitative comparison is required. Researchers commonly prepare cell lysates in RIPA buffer with protease inhibitors, clarify by centrifugation at 1000 x g, and load diluted supernatant against the standard curve, treating each well as a single replicate for downstream analyses. Intended for research use only.
