Human TPH2 ELISA Kit

A Sandwich ELISA Kit from Assay Genie intended for the quantitative measurement of Tryptophan Hydroxylase 2 (TPH2) in human samples. The assay uses a double-antibody sandwich format with colorimetric detection, and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.188 ng/mL, with an assay range of 0.313–20 ng/mL. Reagents are intended to be stored at 2–8°C for up to 12 months, and absorbance is read at 450 nm after TMB substrate development.

TPH2 is a critical enzyme in the biosynthesis of serotonin, a neurotransmitter involved in the regulation of mood, behaviour and cognition, and its dysregulation has been implicated in psychiatric conditions including depression, anxiety and schizophrenia, making it a relevant target in neuroscience research. Linearity has been evaluated in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, and recovery has been characterised across the same matrices. The procedure follows a standard sandwich ELISA workflow: reagent equilibration and plate set-up; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody binding; HRP-streptavidin binding; TMB colour development in the dark; and a stop reaction before reading.
