Human TPH1/Tryptophan Hydroxylase 1 ELISA Kit

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of human TPH1 (Tryptophan Hydroxylase 1). The assay uses a double-antibody format and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. TPH1 is an essential enzyme involved in serotonin biosynthesis, and the kit is intended for research applications in neuroscience relating to mood regulation and behaviour.

Performance characteristics quoted from the manufacturer include a detection range of 0.313-20 ng/mL and a sensitivity of 0.188 ng/mL. Linearity for serum across 1:2, 1:4 and 1:8 dilutions is 86-105%, 87-103% and 85-104% respectively; EDTA plasma gives 83-96%, 84-101% and 84-101%; heparin plasma gives 82-95%, 81-90% and 82-94%. Spike-and-recovery values are 88-101% (average 96%) for serum, 88-99% (average 95%) for EDTA plasma, and 89-101% (average 95%) for heparin plasma. Reagents should be stored at 2-8°C, and absorbance is read at 450 nm.

The procedure follows a standard sandwich workflow: pre-coated plate, primary incubation at 37°C, biotin-labelled detection antibody, HRP-Streptavidin conjugate, TMB substrate development, and a stop step prior to reading.
