Description

A competitive ELISA kit from Assay Genie designed for the quantitative detection of human TPD52 (Tumor protein D52). The assay uses a microplate pre-coated with TPD52 antigen and a biotinylated detection antibody, with HRP-Streptavidin (SABC) and a TMB substrate for colorimetric readout measured at 450 nm. Sample or standard analyte competes with the coated antigen for binding sites on the detection antibody, and TPD52 concentration in the samples is determined by comparing the optical density of the samples to the standard curve.

The kit is validated for use with human samples, including serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Reported performance includes a sensitivity of 0.188 ng/mL and a standard range of 0.313-20 ng/mL. Linearity in serum (n=10) at 1:2, 1:4 and 1:8 dilutions is 86-102%, 93-105% and 85-105% respectively; EDTA plasma (n=10) 86-98%, 90-99%, 86-101%; heparin plasma (n=10) 82-98%, 81-98%, 81-99%. Recovery averages 91% in serum, 95% in EDTA plasma and 95% in heparin plasma. Intra-assay CV is <8% and inter-assay CV is <10%. The associated UniProt entry is P55327, and documented synonyms include TPD52, Tumor protein D52, Protein N8, D52, N8L, PC-1, PrLZ and hD52. TPD52 is described as playing a role in cancer progression and metastasis, making it a candidate biomarker for cancer research.

Kits should be stored sealed at 2-8°C and not cryopreserved, with light-sensitive components protected from direct light. The protocol involves equilibrating TMB substrate for 30 minutes at room temperature, adding 50 µL of standard or sample together with 50 µL of biotin-labelled antibody to each well, incubating at 37°C for 45 minutes, washing the plate five times with wash buffer, developing with TMB substrate in the dark at 37°C for 10-20 minutes, adding stop solution (colour changes from blue to yellow), and reading absorbance at 450 nm. The product is intended for research use only.
