Human TOP2A/Topoisomerase II Alpha ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of human TOP2A (Topoisomerase II Alpha) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody, colorimetric assay has a sensitivity of 0.188 ng/mL and a detection range of 0.313–20 ng/mL.

The protocol follows six steps: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody binding for 60 minutes; HRP-streptavidin binding for 30 minutes; TMB substrate colour development in the dark for 10–20 minutes; and a stop reaction read at 450 nm. Linearity in serum (n=5) at 1:2, 1:4 and 1:8 dilutions is 81-100%, 87-96% and 82-98%; EDTA plasma is 82-98%, 87-100% and 83-101%; heparin plasma is 84-99%, 83-92% and 88-95%. Recovery ranges average 95% for serum (85-101%), 93% for EDTA plasma (85-105%) and 93% for heparin plasma (85-100%). Kits should be stored at 2-8°C for 12 months.
